Earle’s Balanced Salts Solutions (EBSS) are chemically defined, preserving the integrity of mammalian cells in vitro. Designed for short-term use in a CO2 environment, EBSS maintains osmotic pressure and physiological pH (7.2-7.6), and supplies essential metabolic components. Available with or without Calcium or Magnesium and Sodium Bicarbonate, a specialized version without calcium and magnesium is used during cell dissociation to prevent trypsin inhibition.

Chemically defined Balanced Salt Solutions conserve the structural and physiological integrity of mammalian cells in vitro by maintaining intra and extra-cellular osmotic pressure, providing essential components such as glucose, water, and inorganic ions for cellular metabolism, and preserving physiological pH conditions (7.2-7.6).

HEPES is used in many media because it has more buffering capacity than sodium bicarbonate at physiological pH (7.2 – 7.4) at 37°C. Sodium bicarbonate is nutritionally necessary for most cells, so HEPES should be added in addition to, not in place of, sodium bicarbonate.
